Empty FPLC Column Introduction

Empty FPLC columns are specifically designed for applications such as antigen-antibody purification, recombinant protein purification, and protein desalting. According to your needs, you can fill the columns with chromatography media designed for different separation mechanisms, including immunoaffinity, ion exchange, gel filtration, and reverse phase chromatography. These columns are powerful tools for purifying antibodies, proteins, and other molecules. The empty FPLC columns are compatible with liquid chromatography systems (e.g., ÄKTA purifier) and can also be connected to a syringe or peristaltic pump.

Packing Method

1. Frit Preparation
   Use tweezers to gently hold one end of the frit and place it into distilled water or 10% ethanol to prevent air bubbles from remaining in the frit pores. Alternatively, place the frit into a beaker containing distilled water or media storage solution and use an ultrasonic cleaner to sonicate for a few seconds to remove trapped air bubbles.
2. Installing the Bottom Frit
   Take one frit and place it at the top opening of the column. Use the column-packing rod to push it down to the bottom of the column.
3. Filling the Media
   Use a dropper to add the media (resin) drop by drop into the column. Let it stand briefly to allow the media to settle. Once the media has settled to the desired level, carefully add the storage buffer until the column is full.
4. Installing the Top Frit
   Take another frit and place it at the top opening of the column. Use the packing rod to gently push it horizontally to the top of the settled media.
   Note: If the frit tilts inside the column, gently adjust its top edge with a glass rod until level, then use the packing rod to push it to the media’s surface.
5. Installing the Sealing Plug
   Place the column tube into the manual packing tool’s holder, then return the holder to its base. Place the sealing plug flat onto the column’s top end, squeeze the handle firmly to embed the plug into the column. Remove the column from the holder.
6. Sealing and Storage
   Push the sealing sleeve down over the top of the column tube until it snaps into place. Insert the top end cap into the sealing plug and tighten it. Store the packed empty FPLC column at 4°C in a refrigerator.
