Normal Phase HPLC Column
Normal phase high-performance liquid chromatography (HPLC) is a chromatographic technique used for the separation and analysis of compounds that are non-polar or moderately polar. In normal phase HPLC, the stationary phase is polar, while the mobile phase is non-polar. Here are normal phase HPLC columns produced by HAWACH:
| Bonded Phase | USP No. | Particle size (μm) | Pore size | Surface area(m2/g) | Carbon loading | pH Range | End-capped | Price |
| SiO2 | L3 | 5 | 120Å | 320 | N/A | 1.5-10 | No | |
| NH2 | L8 | 5 | 120Å | 320 | N/A | 1.5-10 | No | |
| CN | L10 | 5 | 120Å | 320 | 6% | 1.5-10 | No | |
| Diol | / | 5 | 120Å | 320 | 2.50% | 1.5-10 | No | |
| Amino Acid | / | 5 | 120Å | 320 | 17% | 1.5-10 | Yes |
- Silica-based columns: Silica is the most commonly used stationary phase in normal phase HPLC. It consists of porous particles coated with a thin layer of bonded silica gel. Silica columns offer excellent separation of non-polar compounds but can be sensitive to moisture and require careful conditioning before use.
- Diol-based columns: Diol (dihydroxypropyl) stationary phases are commonly used for normal phase HPLC. These columns contain a polar diol group attached to the silica gel surface. Diol-based columns provide good selectivity for compounds containing polar functional groups such as alcohols, phenols, and amines.
- Cyanopropyl-based columns: Cyanopropyl stationary phases contain a cyano group attached to the silica surface. These columns provide enhanced retention and selectivity for polar compounds, such as compounds with carbonyl groups (e.g., ketones and aldehydes).
- Amino-based columns: Amino stationary phases contain amino groups bonded to the silica surface. These columns are used for the separation of polar compounds, especially those containing acidic functional groups. Amino-based columns can provide strong ionic interactions with acidic compounds.
- Unbonded silica columns: Unbonded silica HPLC columns are made of bare silica particles without any bonded phase. These columns are used for separations based on polar interactions and can be useful for the analysis of very polar compounds.
It’s important to note that normal phase HPLC requires the use of non-polar solvents as the mobile phase, such as hexane, isooctane, or dichloromethane. Additionally, column selection should be based on the specific separation requirements and the properties of the compounds to be analyzed.
