Overview of Chromatographic Separation
What is Chromatographic Separation(CS)?
CS is an effective method of separating various components derived from complicated mixtures. It obeys the matter that different substances possess different distribution coefficients in the system constructed by the stationary phase and mobile phase.
When the two phases perform the relative movement, these substances flow together with the mobile phase and are distributed repeatedly between the phases to separate all targets.
Category of Chromatogram
According to the type of stationary phase and separation principle, chromatograms can be divided into adsorption, partition, ion exchange, affinity, macroporous adsorption resin, gel, and focusing chromatography, among which the most commonly applied is adsorption chromatography.
Here is the adsorption chromatography mentioned first.
It requires the separation of components in a mixture when the mixture along with the mobile phase passes through the adsorbent, different substances are apart due to the fact that adsorbent has different adsorptive power in terms of different materials. This method is especially suitable for the separation of fat-soluble components.
Applications
Chromatography is a very important and commonly used technique in the purification and analysis of biological macromolecules, notably in the purifying and analyzing of proteins.
