An Overview of Centrifuge Tube

As one of the frequently used tools in the lab, the centrifuge tube is able to conveniently and safely samples. For the processing of larger volume samples, conical-bottomed screw cap centrifuge tubes are usually used. Today, HAWACH will mainly discuss the features, technical advantages, classifications, and possible reasons for the rupture of the centrifuge tube during use.

Features of centrifuge tubes

Made of high-quality transparent polymer material polypropylene (PP), it has been widely used in a variety of experimental operations to meet the requirements of bioanalytical grades.
1. 3 types available in HAWACH: flat bottom, V-shaped bottom, and round bottom centrifuge tubes.
2. The sealing cover has a sealing ring, which has good sealing performance.
3. The tube surface, both inner and outer are uniform and smooth.
4. Clear scale is printed.
5. Withstand temperature range: -80℃~121℃.
6. Strictly tested for leaks.

Technical advantages of centrifuge tubes

1. It can effectively deal with some difficult components in soil samples such as eubacterial spores, endospores, Galanz-positive bacteria, yeast, nematodes, seaweed, fungi, etc.
2. The MT buffer and sodium phosphate buffer used in the sample processing process can effectively protect the nucleic acid during the entire extraction process and reduce RNA pollution to a greater extent.
3. The purification method based on silica beads can effectively remove PCR inhibitors such as humic acid and polyphenols
4. The purified DNA can be directly used in subsequent experiments such as PCR and enzyme digestion.
5. For samples with a particularly high humic acid content, additional treatment methods are added.

Centrifuge tube specifications and classification

The function of the centrifuge tube is the container used to hold the separated samples when the centrifuge processes the samples. It is used for the separation of the centrifuge. The centrifuge tube has many specifications. HAWACH will take you to understand it.

1. Centrifuge tubes can be divided into plastic centrifuge tubes, glass centrifuge tubes, and steel centrifuge tubes according to their materials. Commonly used materials for plastic centrifuge tubes are polypropylene (PP), polyethylene (PE), polycarbonate (PC), etc. Its advantage is that it is transparent or translucent, its hardness is small, and the sample can be taken out by the puncture method. The disadvantage is that it is easy to deform, has poor corrosion resistance to organic solutions, and has a short service cycle. Polypropylene (PP) is the best material in plastic centrifuge tubes, so we try to use PP materials when selecting plastic centrifuge tubes.

When using a glass centrifuge tube, the centrifugal force should be reduced as much as possible, and a rubber pad should be used to prevent the centrifuge tube from breaking. High-speed centrifuges generally do not use glass centrifuge tubes.
The steel centrifuge tube has high hardness, no deformation, heat resistance, freezing resistance, and chemical corrosion resistance.

2. According to the capacity of the centrifuge tube, it can be divided into the following categories: microcentrifuge tubes, generally 1.5ml, 2.0ml centrifuge tubes; ordinary centrifuge tubes, generally 15ml, 50ml centrifuge tubes; A large number of centrifuge tubes, generally 250ml, 500ml centrifuge tubes, centrifuge tubes larger than 250ml can also be called centrifuge bottles.

3. According to the shape of the bottom, it can be divided into conical centrifuge tubes, round-bottom centrifuge tubes, and flat-bottom centrifuge tubes. Among them, conical centrifuge tubes are commonly used.

4. According to the closing method of the lid, there are gland centrifuge tubes and screw cap centrifuge tubes. The gland type is often used for microcentrifuge tubes, and the screw cap is often used for centrifuge tubes with larger capacity or centrifuge bottles.

Possible problem- rupture of the centrifuge tube during use

Many customers encounter the rupture of the centrifuge tube when they use it at ordinary times. What is the cause? HAWACH will briefly discuss about the reason for the rupture of the centrifuge tube during use.

1. The sample placed in the symmetrical sleeve hole must be balanced with the laboratory centrifuge tube together with the balance to adjust the weight. If only one test tube is used to put the sample, the symmetrical test tube must be balanced by adding water, otherwise, the instrument will vibrate during centrifugation and the test tube will be broken. What is easily overlooked is that the centrifuge cup of the horizontal rotor will be unbalanced after long-term use or replacement, so the centrifuge cup should also be balanced. For example, the failure of the test tube of a centrifuge that has been used for more than 10 years is difficult to eliminate. Later, it was found that the four centrifuge cups were overweight, and the failure disappeared after the cups were replaced.
2. The speed of the centrifuge is too high, and the centrifugal force that exceeds the centrifugal force that the centrifugal tube can withstand will also break the test tube.
3. Centrifugating too fast may also be the reason to break the laboratory centrifuge tube.
